Effective capturing chloroplast RNA through poly(mA) tailing for nanopore direct total RNA sequencing

We developed an efficient 3 end custom tailing method to sequence both poly(A) and non-poly(A) RNAs derived from Populus leaf tissue. We assessed this method through sequencing in vitro transcribe RNA tailed with ATP or modified ATP analogs. We further tested this method with natural cellular RNAs from P. trichocarpa leaf tissue.
