Since the outbreak of avian adenovirus in multiple provinces of China in 2015, it has caused huge losses to the poultry industry. After infection in chicken flocks, various clinical symptoms such as pericardial effusion hepatitis syndrome, inclusion body hepatitis, and myogastric erosion can be caused. The disease spreads in various ways, has no obvious seasonal pattern, and has a high mortality rate (30% to 80%). These hazards have put forward more requirements for the biological safety and control level of various chicken farms. How to protect the health of chicken populations and ensure the normal production of chicken farms is a key focus of attention for chicken farming enterprises and related research institutions. This study compared and analyzed the gene sequences of 12 serotype strains of avian adenovirus, designed fluorescent probe primers at conserved genes, optimized the amount of fluorescent probe primers added to the reaction system, and simultaneously detected 161 clinical samples with industry standards. A multi serotype and multi TaqMan fluorescence quantitative PCR detection dataset for avian adenovirus was established. Specific detection of FAdV-4, FAdV-8a, and FAdV-8b showed no cross reactivity with other serotypes of avian adenovirus, NDV, AIV H9 subtypes, IBV, etc; The minimum detection value is 17 copies/μ L; The coefficient of variation (CV%) within and between groups ranges from 0.1% to 1.1%. The overall positive compliance rate with industry standards is 100.00%. This research achievement not only detects three pathogens in a single tube and can be used for epidemiological investigation and early rapid screening of avian adenovirus diseases in chicken farms, but also lays the foundation for theoretical research and practical application of biosecurity prevention and control technologies for other diseases in chicken farms.
