To determine the rhythmic expression of genes potentially involved in regulating physiological and circadian rhythms in P. xylostella, we performed RNA-seq analysis on the head samples of P. xylostella collected every 4 hours. The G88(WT), Pxper-KO(M10), Pxcry2-KO(1.1) and PxperPxcry2-dKO(3.1) were subjected to a 4-day light-dark (14hL:10hD) cycle of 14L:10D. Male adult heads were collected at LD4 time points for RNA extraction, with two independent replicates per time point. Paired-end RNA-seq (2 x150 bp) sequencing strategy was performed using the Illumina NovaSeq 6 000 sequencer.
