SMuRF was developed with the aim of creating a universal deep mutational scanning workflow. Initially, it was established to characterize all possible coding SNVs for FKRP, and subsequently, its adaptability was demonstrated by its application to LARGE1. Amplicons from each block of each gene of both the high-alpha-dystroglycan group and the low-alpha-dystroglycan group were multiplexed together for sequencing. Samples from the high groups are barcoded with "tACACGATC" and low "AAGTAGAG". Essentially, SMuRF score is the normalized relative high/low enrichment of a variant in the FACS groups.
