The goal of this study is to examine how maternal diet exposure (nornal control diet vs. high-fat-deitor low-protein diet) impact adult beta-cell function and gene expression. Mice were used as models. Overall design: Female wild-type mice (CD1) (~7 wks after birth) were fed with control diet, high-fat-diet or low-protein diet for two weeks. They were then crossed with Ins1Apple males. After birth, all mice were switched to control diet until tissue collection. Islets were isolated ~ 7 wks after birth, dissociated, and FACS-sorted for Total RNA exaction. Totl RNA were prepared using a DNA-free RNA kit from Zymo, quality-checked with Agilent Pico RNA analyzer, and suqenced using the Illumina platforms (NovaSeq 6000) in the Vanderbilt Technologies for Advanced Genomics (VANTAGE). VANTAGE also provided quality control data and primary data processing for alignment and feature (gene) identification. Only RNA preps with RIN>=9.4 were used for these experiments.
