Single cell RNA sequencing of placenta in patients with early onset preeclampsia

Early-onset preeclampsia (EOPE) is a severe hypertensive disorder of pregnancy, frequently associated with fetal growth restriction (FGR) and reduced estrogen levels due to placental dysfunction. We employed single-cell RNA sequencing (scRNA-seq) to profile placental trophoblasts from EOPE patients complicated by FGR. Overall design: After collecting placental tissues, they were immediately processed into single-cell suspensions. The process involved washing, cutting, digesting, filtering, lysing red blood cells, removing dead cells, staining, and finally suspending the cells in a solution. The single-cell suspension was loaded onto the Chromium Controller to generate single-cell gel beads, followed by single-cell RNA sequencing using the Illumina Novaseq6000 sequencer.
