The development of new strategies for articular cartilage regeneration has been investigated, including the use of biomolecules secreted by mesenchymal stem cells (MSCs). We have developed a culture platform based on the use of specific bioreactors for the large-scale culture of from human dental pulp MSCs (hDPSCs) under chondrogenic stimuli. We have optimized secretome production based on known chondral markers (TGF-β1, SERPINE1, and the miRNAs miR-23a, miR-204, miR-211 and miR-337-5p) and have characterized its composition at the metabolomic and proteomic level. We have also studied the chondrogenic potential of the secretome using representative <i>in vitro</i> models. In this study, we developed an MSCs culture system that allows for the production of large volumes of secretome with controlled composition and chondrogenic induction potential. This is a preclinical study necessary for subsequent analysis of the secretome obtained using <i>in vivo</i> experimental models.
