Identification_of_Pyocin_L1_sensitivity_determinants_by_TraDIS

Pyocin L1 is a lectin-like bacteriocin produced by Pseudomonas aeruginosa, which displays genus-specific killing activity against Pseudomonas. Pyocin L1 binds to D-rhamnose and this is a prerequisite for its bactericial activity. However, its mechanism of cell entry and eventual killing is not fully understood. Here, we have used a genome-wide screening approach to define the set of genes which, when mutated, result in increased or decreased susceptibility to killing by Pyocin L1. We performed TraDIS on a saturated mutant library of P. aeruginosa PAO1 with and without Pyocin L1 challenge, and have identified multiple genes that affect Pyocin L1 susceptibility.
