In this study, PHB was characterized for the first time to be a RNA binding protein its role in bladder cancer aggressiveness was investigated. Anti-PHB RIP-SEQ of two paired of bladder cancer tissues from both tumor tissues and normal tissues identified greater binding of NADSYN1 mRNA and PHB specially in tumor tissues compared with adjacent normal tissues. RIP-SEQ assays were performed with a Magna RNA-binding protein immunoprecipitation kit. Negative control IgG, human anti-PHB antibody were used in this study. Protein-RNA complex were purified and immunoprecipitated RNAs were extracted after proteinase K digestion. Purified RNAs were then subjected to RNA-SEQ.
