Exome and transcriptome analysis of implanted MC38 WT or MC38 PD-L1-KO tumor tissues for neoantigens identification. Mus musculus

To identify tumor neoantigens, MC38 WT or MC38 PD-L1-KO cells were inoculated into C57BL/6J mice. 2 weeks after inoculation, tumor tissues were resected and divided into 2 tubes for DNA or mRNA extraction. Whole blood cells from non tumor bearing mice were collected for DNA extraction as germline control. Exome or transcriptome sequence library were prepared by using SureSelect Mouse All Exon Kit (Agilent Technologies) or SureSelect Strand-Specific RNA Library Prep Kit (Agilent Technologies), respectively. mRNA extracted from neoantigen specific T cells identified in this study were further processed by using SMARTer Mouse TCR a/b Profiling Kit (Clonetech) for library preparation.
