Purpose: To compare the TnAV-6a1 variant infected Trichoplusia ni larval transcriptome of hemolymph viral vesicles versus those of other major larval tissues specifically, the fat body, epidermis and tracheal matrix (referred to as FET tissues). Method: We used a TnAV-6a minor variant strain (referred to as TnAV-6a1) to infect third or early fourth instars Trichoplusia ni larvae. Then, collected the hemolymph viral vesicles and fat body, epidermis and tracheal matrix for transcriptomic studies.The TnAV-6a1 variant genomic DNA library prepared following manufacturer instructions of the NEBNext DNA Library Prep Master Mix Set for Illumina (New England, BioLabs). Thirty four contig sequences were generated using Trinity software with “genome guided” assembly option (Grabherr et al., 2011). For genome sequencing we used the Illumina MiSeq (2 x 300, Paired-end). For RNA-sequencing we used the HiSeq2500 Illumina sequencer (Noll Core Facility at the UC Riverside Institute for Integrative Genome Biology), in duplicate.
