Transcription Factor SOX30 Is a Key Regulator of Mouse Spermiogenesis

As Sox30 knockout arrests spermatogenesis at the step 3 round spermatid (rST) stage, we performed RNA sequencing to identify genes regulated by Sox30. Overall design: We conducted RNA-sequencing (RNA-seq) analysis to identify SOX30-regulated genes that were differentially expressed in rST of Sox30 KO and WT mice. rST were isolated by FACS from 23 dpp (days postpartum) WT and KO mice, a stage where spermatogenesis progress to step 3 rST with no extra haploid cell types present.
