Canine lung tumors may belong to canine primary lung cancer or caused by the metastasis of malignant tumors from other tissues and organs. Due to the unclear molecular mechanism, the research and use of small molecule drugs and immune checkpoint inhibitor drugs are affected. In recent years, high-throughput sequencing technology has been widely used in canine cancer research, laying a foundation for the study of cancer-related pathogenic mechanisms. However, there are no reports on the research of single-cell RNA sequencing (scRNA-seq) of canine lung tumors. In this study, tumor and para-tumor samples were obtained from the lungs of a suspected canine primary lung cancer case, and then the samples were analyzed by single-cell sequencing. Sequencing analysed a total of 18518 cells and identified 8 cell types, include Epithelial cells, Endothelial cells, Fibroblasts, Smooth muscle cells, Macrophages, Monocytes, T cells, B cells and two unclassified clusters. Through GSVA, GO, DEGs and cell-cell interaction analysis, the formation of the tumor microenvironment in the samples and its influence on the and development of tumors were described, furthermore, according to the molecular features presented by the sequenced samples, targeted drugs for potential testing were also proposed. As canine primary lung cancer has similar clinical characteristics to human lung cancer, this research can not only deepen our understanding of the cellular and molecular characteristics that affect tumor progression in canine lung tumors, facilitate the diagnosis and treatment of canine lung cancer, but also provide data references for studying human lung cancer when using canine lung tumor as a model.
