Background At present, the treatment of chronic prostatitis (CP) has not achieved the expected efficacy, so it is urgent to find new treatment drugs. The preliminary clinical observation of the research group found that Anemone Tomentosa has a good effect on chronic prostatitis, but the specific mechanism of action remains to be further studied. Objectives To investigate the role of prostasome derived miRNA-146a in regulating TLR-4/NF-κB pathway in chronic inflammation in EAP rats and the mechanism of Anemone Tomentosa in treating EAP rats. Methods Forty-two Wistar rats were divided into 7 groups by random number table method, including normal group, model group, mimics group, inhibitor group, low-dose of anemone tomentosa group, medium-dose of anemone tomentosa group and high-dose of anemone tomentosa group, with 6 rats in each group.After drug intervention, miRNA-146a-5p mRNA in prostasome will be detected by RT-PCR, and TLR-4, IκB, p-IκB and TRAF6 protein will be detected by WB, and inflammatory cytokines include TNF-α, IL-6, IL-8, and IFN-γ will be detected by ELISA. Results Compared with normal group, miRNA-146a-5p mRNAs expression in prostasome were down-regulated in each group (P<0.01). Compared with model group, miRNA-146a-5p mRNAs expression in prostasome were up-regulated in medium-dose group, high-dose group, and mimics group (P<0.01), and were down-regulated in inhibitor group(P<0.01).The expression of TLR-4, p-IκBα and TRAF6 proteins in prostate tissues of EAP rats were significantly down-regulated in anemone tomentosa and mimics groups (P<0.01).The expression of IκBα proteins in prostate tissues of EAP rats were significantly up-regulated in anemone tomentosa and mimics groups(P<0.01).The serum levels of TNF-α, IL-6, IL-8 and IFN-γ in EAP rats were significantly down-regulated in anemone tomentosa and mimics groups(P<0.01or P<0.05).Anemone tomentosa and miRNA-146a mimics significantly reduced the inflammatory cell infiltration of prostate tissue and improved the pathological injury of prostate tissue. Conclusion Prostasome derived miRNA-146a can regulate the activation of TLR-4/NF-κB pathway participate in local pathological changes of prostate in EAP rats.The mechanism of the anemone tomentosa inhibiting chronic inflammation in EAP rats may be related to the regulation of prostasome derived miRNA-146a/TLR-4/NF-κB pathway.
