This study aims to enhance malaria molecular surveillance by simplifying and expanding the PfSMARRT (24 amplicons) assay into a more accessible and scalable platform, termed PfSMARRTer. The primary goal is to support real-time tracking of Plasmodium falciparum drug resistance by improving the detection of key genetic mutations associated with artemisinin partial resistance and resistance to partner drugs. By streamlining the library preparation protocol, eliminating the need for DNA normalization, and introducing additional validated resistance markers, PfSMARRTer significantly lowers the technical and cost barriers that limit the adoption of highly multiplexed sequencing in endemic regions. Its open-source, automation-friendly design is especially relevant for resource-limited settings, where timely detection of emerging resistance is critical for informing treatment policies and preventing the spread of resistant malaria strains.
