Data used to demonstrate confocal reflectance imaging as a highly sensitive imaging modality for 3D cell culture. Data includes reflectance and fluorescence imaging of cultured osteocyte and fibroblast cells in 3D polyisocyanopeptide (PIC) hydrogel, as well as collagen, fibrin, and Matrigel. Confocal reflectance imaging typically suffers high background and poor sensitivity. We demonstrate sensitive and low-background reflectance imaging of cells encapsulated in transparent 3D hydrogels. Nanoscale cell morphology is visualized with sensitivity similar to confocal fluorescence, with low laser power, minimal specimen preparation, and reduced toxicity.
