Cordyceps sinensis is a complex of insects and fungi, possessing various pharmacological activities. As a precious Chinese herbal medicine, it has a unique growth environment and life cycle, and it needs to be induced by low temperature to form fruiting bodies. At present, the mechanism by which low temperature affects the growth and development of Cordyceps sinensis remains unclear, and there is a lack of research data. This thesis takes the Cordyceps sinensis monocospores strain TZ8-1 from the alpine meadows at the eastern foot of the Qilian Mountains as the research object. After being cultured at normal temperature and low temperature for 10 days, 30 days, 50 days, and 70 days, the morphology of colonies and mycelium, the changes in fresh and dry weight of colonies, and the development of conidia were observed using an optical microscope. Analyze the accumulation changes of key metabolites (such as urea, mannitol, polyphenols and polysaccharides); The RNA-seq and gene expression difference techniques were used to explore the response genes of Cordyceps sinensis under low temperature stress, and to preliminarily investigate the response characteristics of Cordyceps sinensis mycelium to low temperature in terms of appearance morphology, physiology and biochemistry, and gene expression levels, providing a theoretical basis for the development of Cordyceps sinensis fruiting bodies induced by low temperature.
