To examine transcriptional changes with polyploidization in Brachypodium species, we analyzed diurnal transcruiptome in Brachypodium distachyon Bd21, B. stacei ABR114, B. hybridum Bd14-1, and artificially induced tetraploid Bd21. We performed RNA-seq analysis of their leaves sampled through diurnal cycle of over 24-h at 4-h intervals using three biological replications. To reveal the diurnal changes of the transcriptome in Brachypodium distachyon Bd21 and B. hybridum Bd14-1, we performed RNA-seq analysis of its leaves sampled through a diurnal cycle of over 48-h at 4-h intervals using three biological replications. We sampled leaves from Bd21 and Bd14-1 plants grown in a growth chamber for 3-4 days after synchronized germination. To analyze gene expression change in Brachypodium Bd21-3 overexpressing Bradi3g56490, we performed RNA-seq analysis of leaves sampled from an overexpression line and wild type Bd21-3.
