This Zenodo repository contains the R scripts, processed datasets, supporting files, and source data used for the study: "Genetically predicted leukocyte telomere length and susceptibility to sporadic vestibular schwannoma: a two-sample Mendelian randomization study." This study evaluated whether genetically predicted leukocyte telomere length (LTL), insulin-like growth factor 1 (IGF-1), circulating C-reactive protein (CRP), and adult height were associated with susceptibility to sporadic vestibular schwannoma (VS) using a two-sample Mendelian randomization framework. The repository includes: 1. R scripts used for:- GWAS data processing and harmonization;- genetic instrument selection;- Mendelian randomization analyses;- sensitivity analyses;- heterogeneity and horizontal pleiotropy assessment;- MR-PRESSO analysis;- generation of manuscript tables and figures. 2. Processed analysis files, including:- harmonized exposure-outcome datasets;- final leukocyte telomere length instrument datasets;- instrument selection audit files;- sensitivity analysis datasets. 3. Source data supporting manuscript outputs, including:- manuscript tables;- figure source data;- final analysis summaries. 4. Supporting documentation describing:- data sources;- analysis workflow;- file descriptions. Publicly available GWAS summary statistics were obtained from:- NHGRI-EBI GWAS Catalog (GCST90134433 and GCST009856);- IEU OpenGWAS database (ieu-b-35);- GIANT Consortium/Yengo 2022 height GWAS resource;- UK Biobank-based molecular trait GWAS reported by Sinnott-Armstrong et al. The original large-scale GWAS summary statistics are not redistributed in this repository because they are publicly available through the original databases. Instead, processed datasets and analysis files required for reproduction of the reported analyses are provided. The R scripts were developed iteratively during the analytical process. The final manuscript results were generated using the final corrected analysis scripts, while earlier scripts are retained to document the analytical workflow and validation process. The repository was created to improve transparency and facilitate reproduction of the reported analyses.
