Video of fluorescence microspectroscopy based in vitro characterisation of DmCRY-CT / FAD binding and aggregation. 100 μM DmCRY-CT solution with 10 μM FAD (pH 7.4) leads to emissive particle formation and aggregation. DmCRY-CT / FAD aggregation occurs immediately upon imaging but irradiation (450 nm) results in greater particle number and size. Left panel: fluorescence imaging reveals greater accumulation of particles under illumination. Right panel: bright-field microscopy (contrast adjusted to expose the region surrounding the focussed laser irradiation spot) reveals gradual particle accumulation in the area surrounding the irradiation spot (excitation intensity = 2.1 kW/cm2, timescale = 179 minutes).
