Naïve CD4 T cells from healthy adult individuals were isolated using EasySep Human Naive CD4 T cell Enrichment Kit (StemCell Technologies). Cells were transfected with control siRNA (Dharmacon: D-001910-01) or YY1 siRNA (Dharmacon A-011796-16) using the P3 Primary cell 4D-Nucleofector kit and the Lonza 4D-Nucleofector system. Transfected cells were cultured without stimulation for 48 hours, when YY1 protein expression had declined by >50%. Libraries were prepared using the Ovation RNA-seq whole blood kit (NuGen) and sequenced on an Illumina HiSeq 2500.
