Highly regenerative organisms like axolotls initiate a DNA damage response upon injury. To test whether this response is important for the regeneration process, we used CRISPR/Cas9 to target a DNA repair-associated gene: eya2. By doing so we generated Eya2 mutant axolotls that experienced deregulation of their DNA damage response. Our data provide evidence that up-regulation of DNA damage/repair genes during regeneration, including eya2, are important for regeneration to promote cell cycle progression. Samples in this submission reflect individual eya2 animals with different genotypes (wt or mutant) at different time points following limb amputation.
