Massive parallel RNAi knockdown was induced in bloodstream-form Trypanosoma brucei. Flow cytometry was then used to divide the perturbed population according to cell cycle phase and RNAi target fragments were deep-sequenced from each pool using RIT-seq such that flow cytometry profiles could be digitally reconstructed at a genomic scale.
