Modulators of temozolomide response in glioma cells

The alkylating agent Temozolomide (TMZ) is a standard compound in glioblastoma treatment. Most patient develop resistance, however. Furthermore, the therapeutic benefit is limited in those with an unmethylated MGMT promoter. The identification of synthetic lethal interactors thus represents a relevant question to improve therapeutic efficacy.We performed genome-wide CRISPR-Cas9 knockout screens in O6-methylguanine-DNA methyltransferase (MGMT)-unmethylated glioma cell lines under TMZ treatment.
