EBF1 and HIF1a ChIPseq in MDA-MB-157 cells under normoxia and hypoxia Overall design: Cells were culutre under normoxia or hypoxia for 24h, then fixed with 1% formaldehyde for 10 min at room temprature. Fixiation was quenched with 125 mM Glycine. Chromatin was fragmented by Micrococcal nuclease (MNase) digestion. Transcription factor bound chromatin was pulled down using specific antibodies and protein-G megnetic beads. Enriched chromatin then was further de-crosslinked and used for libraries construction.
