This study analyzed the effect of RBM5 gene deletion in cortical brain tissue on differential gene expression/splicing changes 48h after a traumatic brain injury (TBI). TBI was induced in WT vs. KO mice by controlled cortical impact (CCI) injury. The four grouops included: (1) Sham-WT, (2) CCI-WT, (3) Sham-KO, and (4) CCI-KO. The objective of this study was to test if RBM5 KO decreased the expression of cell death mediators in the contused brain 48h post-injury. Overall design: A total of 28 brain tissues were isolated from 12-15wk old mice for microarray analysis. Seven animals were compared per group (n=7/group). Each group contained a mix of female and male mice as indicated for each sample in the list below. Injured mice recieved a craniotomy over the left parietal cortex via a dental drill, and subjected to a CCI injury (6m/s; 1.2mm depth). Control (sham) mice received anesthesia but did not get a craniotomy. 48h after the injury mice were euthanized and cortical tissues collected for total RNA extraction. Extracted RNA was hybridized to microarray chips (MTA 1.0).
