Gene expression data using RNA-seq in central airways of alternaria-exposed allergic asthma mice

This data was generated from total mRNA samples from trachea and central bronchi among alternaria-induced allergic asthma mice model. Female 7-week-old BALB/c mice (n = 3) were received an intranasal instillation 20 uL of 50 ug alternaria extract which was re-suspended in filtered phsphate-buffer saline (PBS) as an asthma mice model. Control mice (n = 3) received 20 uL sterile PBS. Each mouse received the intranasal administration twice a week (Monday and Thursday) for 6 weeks. All mice were sacrificed for evaluation after 24 hours of the last exposure. Trachea and central bronchi were resected and immediately immersed with RNAlater after sacrifice. Total RNA was extracted using RNeasy-Mini spin column as manufacture's instruction. RNA concentration and quality were analyzed using Nanodrop, 18S rRNA/28S rRNA ratio on agarose gel electrophoresis, and RNA Integrity Number (RIN) using 2200 TapeStation (Agilent). Sequencing was performed using NovaSeq 6000 (Illumina). Reference genome sequence for mapping was gencode GRCm38.p6. Expression in each sample was statistically analyzed and was normalized using transcripts per million (TPM) algorithms.
