ssDNA strands of the form ACAC NNNNNNNN ACAC are exposed to various doses of 266nm radiation. The resulting samples are prepared for sequencing using commercial sequencing kits (Swift Accel-NGS 1S) and read out via Hi-Seq (Illumina). Data is used to infer sequence-dependent UV damage yields in high throughput and across a wide range of doses.
