The influence of the heterochromatic marks H3K36me3 and H3K27me3 was elucidated by deletion and (cross-)complementation experiments. Gene expression was analyzed under axenic conditions and in planta by RNA-seq, and epigenetic profiles were assessed by ChIP-seq. Overall design: Axenic cultivation F. oxysporum: NO3 2 days; in planta: 6 DPI. Axenic cultivation F. graminearum: ICI+60 mM glutamine 3 days. Material was subjected to RNA extraction for RNA-seq, and was crosslinked with formaldehyde prior to ChIP-seq using indicated antibodies.
