Establishing a Model Composed of Immune-Related Gene-Modules to Predict Tumor Immunotherapy Response

The tumor immune microenvironment plays a significant role in the response to tumor immunotherapy. We utilized a mouse colon cancer tumor model and administered anti-PD-1 therapy, chemotherapy, and combination therapy separately. We collected mouse tumor tissues for gene sequencing to analyze the differences in the tumor immune microenvironment under different treatment backgrounds.Mouse were subcutaneously injected in the flank with 5X10 5 MC38 colon cancer cells. Tumor sizes were measured every 3 days. Tumor-bearing mouse were randomly grouped and intraperitoneally injected with corresponding drugs. Among the drugs, mouse received Oxaliplatin at the dosage of 10 mg per kg once, or were treated with anti-mice PD-1 antibody at the dosage of 250 ug every 3 days for a total of three times. All the mouse whose largest diameter of tumor reached 2.0 cm were sacrificed in accordance with standards for humane treatment. All tumor tissues were collected on day 9, and gene sequencing was performed.
