This study investigates the impact on the transcriptome of lymphatic endotheial cells of a 5bp deletion (GATA2 binding site) in an enhancer element 11kb upstream of the Prox1 gene. Overall design: Comparison of RNA from deletion mutant and wildtype lymphatic endothelial cells isolated directly from embryos at E14.5. Also, to assess haematopoietic potential, comparison of the same samples after 7d co-culture on OP9 feeder cells.
