There is much demand for more water-soluble Mincle ligands which is promissing adjuvants. We synthesized water-soluble Mincle ligad, PEG-TGL, and assessed its potential on adjuvanticity. Comparing to TDB, known ligand for Mincle, PEG-TGL induced less IL-1beta secretion. To elucidate the gene expression differenses leads to IL-1beta secretion, we performed RNA-seq analysis using bone marrow-derived dendritic cells, and revealed that PEG-TGL were less able to active phagocytic pathways.
