We will use the recently completed rhesus macaque genome map, to identify at least 150,000 candidate single nucleotide polymorphisms (SNPs) randomly distrubuted throughout the rhesus (Macaca mulatta) and longtail (M. fascicularis) macaque genomes and use these SNPs to chatracterize the population structure of rhesus and longtail macaques. DNA fragments of pyrosequenced DNA samples from two different rhesus macaques and one longtail macaque will be compared with the rhesus genome map to identify SNPs using bioinformatics tools we have already developed. A sufficient number of these SNPs (16,720) to identify at least 10,000 of the most informative SNPs in each of the two species will be genotyped and their frequencies estimated in geogrqaphically representative samples of 96 Indian and Chinese rhesus macaques and 96 longtail macaques that also includes several multi-generation families for confirming expected patterns of Mendelian segregation of candidate SNPs.
