7-day-old seedlings grown on MS plates were transferred to small Petri dishes containing 15 mL of liquid 1/2 MS medium supplemented with DMSO or 0.6 mM cordycepin and incubated for 2, 4, 6, 8, 10, and 12 hours. mRNAs were enriched using oligo(dT)-conjugated magnetic beads, and strand-specific libraries were prepared and sequenced on the Illumina HiSeq 2500 platform using a PE150 strategy.
