To identify origins of replication in Lachancea waltii in order to perform a comparative analysis of replication origins between L. waltii and Saccharomyces cerevisiae. Origins of replication in L. waltii were finely mapped through an autonomously replication sequence (ARS) assay. This assay was accomplished by screening a genomic library for L. waltii for ARS activity. The pre-screened and screened libraries were sequenced by Illumina sequencing. Using the list of identified origins, we compare the sequence, nucleosome composition, and location of origins between L. waltii and S. cerevisiae to understand how origins are conserved over evolutionary time.
