This study investigates the impact of splenectomy on the cardiac immune cell landscape post-I/R injury. We performed single-cell RNA sequencing (scRNA-seq) on CD45+ immune cells isolated from mouse hearts 24 hours after I/R injury in both splenectomized and sham-operated (control) mice. Male C57BL/6 mice aged 8 weeks were randomly assigned to two groups: splenectomy and sham surgery. both groups underwent myocardial ischemia-reperfusion (I/R) injury induced by occluding the left anterior descending (LAD) coronary artery for 60 minutes, followed by 24 hours of reperfusion. Hearts were then harvested, and CD45⁺ immune cells were isolated using magnetic-activated cell sorting (MACS).
