Gene expression profile at single cell level of total peritoneal cells from WT and HDC KO mouse

Histamine is required for full differentiation of macrophage. We used single cell RNA sequencing(sc-RNA seq) to characterize the impairment of macrophage differentiation in HDC KO at molecular level. The total peritoneal cells from mice were isolated using 10-ml syringe with ice cold PBS, and cells were prepared for scRNA-seq on a LUNA-FL Automated Fluorescence Cell Counter组胺（Histamine）是巨噬细胞（macrophage）完全分化所必需的物质。本研究采用单细胞RNA测序（single cell RNA sequencing, scRNA-seq），在分子水平上表征HDC敲除（HDC KO）小鼠的巨噬细胞分化受损状态。我们使用10mL注射器配合冰预冷的磷酸盐缓冲液（PBS）分离小鼠腹腔总细胞，并通过LUNA-FL全自动荧光细胞计数器完成适配scRNA-seq的细胞制备工作。
