Histamine is required for full differentiation of macrophage. We used single cell RNA sequencing(sc-RNA seq) to characterize the impairment of macrophage differentiation in HDC KO at molecular level. The total peritoneal cells from mice were isolated using 10-ml syringe with ice cold PBS, and cells were prepared for scRNA-seq on a LUNA-FL Automated Fluorescence Cell Counter
组胺(Histamine)是巨噬细胞(macrophage)完全分化所必需的物质。本研究采用单细胞RNA测序(single cell RNA sequencing, scRNA-seq),在分子水平上表征HDC敲除(HDC KO)小鼠的巨噬细胞分化受损状态。我们使用10mL注射器配合冰预冷的磷酸盐缓冲液(PBS)分离小鼠腹腔总细胞,并通过LUNA-FL全自动荧光细胞计数器完成适配scRNA-seq的细胞制备工作。
