To determine the effects of SARS-CoV-2 infection on the host transcriptional profile, total RNA was isolated from infected cells and analyzed by microarray. For these studies, we took into account that the SARS-CoV-2 replication cycle in cell culture is 6-16 h, followed by death of infected cells 28. Therefore, to evaluate the direct effects of virus infection and avoid the analysis of secondary infection events or cell death effects, we did not extend our analysis beyond 24 h post infection. Analysis of significantly differentially expressed genes showed substantial transcriptomic changes in Calu-3 cells with a total of 3215 differentially expressed genes (FDR< 10%). 2 cell lines (A549-ACE2 and Calu3, 4 time point 4,8,12,24 hours, mock treated vs SARS-CoV2-infected
