We analyzed the effect of RSL3 on T. gondii transcriptome. Principal components analysis (PCA) scores plot and unsupervised hierarchical clustering showed clear separation between parasites treated with RSL3, vehicle 0.1% DMSO and negative control (Ctrl). There were 2,280 up-regulated and 2,342 down-regulated genes identified between RSL3-treated parasites and 0.1% DMSO-treated parasites.. To better understand the biological function of these DEGs, Gene Ontology (GO) enrichment and KEGG pathway analysis were performed. The ‘antioxidant activity’ and cell growth and death’ pathways were significantly downregulated, indicating that RSL3 disrupted redox homeostasis in T. gondii.
