Escherichia coli O157:H7 is one of the major foodborne pathogens in the United States. Previous studies demonstrated that cattle can be a reservoir of E. coli O157:H7 cells. The E. coli O157:H7 strain C1-057 was isolated from finishing cattle in a commercial feedlot. The strain C1-057 was characterized by a multiplex PCR assay to detect the following genes, rfbE (encoding O157 antigen) fliCh7 (encoding H7 antigen), eae (encoding intimin), stx1 (encoding Shiga toxin 1) and stx2 (encoding Shiga toxin 2). The PCR results confirmed that the strain C1-057 carried the rfbE, fliCh7, eae, but neither stx1 nor stx2. Whole genome sequencing was performed by Pacbio RSII. Raw reads were then de novo assembled into a complete genome using SMRT analysis software (v2.3.0). This project was funded by National Cattlemen's Beef Association.
