We studied effects of the mouse sst1 locus that controls susceptibility to intracellular bacteria, on macrophage response to TNF. Bone marrow-derived macrophages were isolated from the B6 wild type and the sst1 congenic B.Sst1S mice and differentiated in vitro in the presence of CSF1. Cells were stimulated with 10 ng/ml of TNF for 12 hours prior to RNA isolation.
