Analysis of the functional significance of peptide-N(4)-(N-acetyl-beta-glucosaminyl)asparagine amidase in human melanoma cells. The hypothesis tested in the present study was that this glycosidase may be involved in melanoma development. Short hairpin RNAs were used to knockdown the expression of peptide-N(4)-(N-acetyl-beta-glucosaminyl)-asparagine amidase in human melanoma cells. The total RNA sample were isolated from the cell samples and prepared for global gene expression profiling using HT-12v4 Human Gene Expression Bead Chips and a HiScan array scanning system. The study included 12 experimental conditions. Each experimental condition had two biological replicates, for a total of 24 samples in the analysis. Samples with non-targeting shRNA (NT-shRNA) served as controls.
