Transcriptomes analysis of long noncoding RNA (lncRNA) and mRNA expression profiles of the porcine alveolar macrophages (PAMs) after porcine reproductive and respiratory syndrome virus (PRRSV) infection in vitro. We obtained 105,627,026 clean reads from 109,443,286 raw reads. A total of 951 annotated and 751 novel lncRNAs were identified. PAMs showed distinct transcriptome profiles after PRRSV infection. It was observed that 126 lncRNAs and 753 mRNAs were differentially expressed between PRRSV-infected and control group PAMs. Overall design: PAMs from the six pigs were divided into six groups (1×107 cells per group), three groups were infected with PRRSV-WUH3 at a multiplicity of infection (MOI) of 0.1, and the other three groups were treated separately with RPMI-1640 culture medium separately. At 30 h after treatment, cells were washed twice with PBS and harvested by scrapers and centrifugation. Aftrt RNA extraction, six RNA libraries (PRRSV1,2,3 and Mock1,2,3) of PAMs were sequenced on a Hiseq2500 platform.
