The abnormal communication between the hematopoietic system and the skeletal system cells is an important cause of imbalanced bone remodeling in the elderly, but the mechanism of this communication abnormality is unclear. Here, we found that knockout of the CLC-3 chloride channel accelerates early bone formation and mid-late bone resorption through microCT analysis. Single-cell sequencing and Spatial Transcriptomics were performed on bone marrow cells from WT and CLC-3-/--/- mice at three representative time points of bone mass changes, 1, 2, and 8 months old, to unravel the mechanism of accelerated bone remodeling imbalance caused by CLC-3 knockout.
