Expression analysis of Alpha-gliadin Genes in Bread Wheat

Characterization of the variants and analysis of the expression profile of an alpha-gliadin amplicon in two low-gluten RNAi wheat lines, with the gliadins strongly down-regulated, and their wild type under two different Nitrogen fertilization conditions. This amplicon encompasses the most immunogenic complex for Coeliac Disease. Expression of this amplicon is reduced in both RNAi lines in comparison to the wild type. Fertilization with high nitrogen regime significantly increases transcripts of the amplicon in the wild type, but not in the RNAi lines. Results of this study will facilitate more comprehensive criteria of alpha-gliadin gene family expression in response to RNAi and nitrogen fertilization.
