Atopic dermatitis (AD) is an inflammatory skin condition characterized in part by elevations in type 2 immune cytokines such as IL-4 and IL-13. Dupilumab is a monoclonal antibody that targets the shared receptor for these cytokines and has shown remarkable efficacy in reducing inflammation in AD. The goal of this study was to study the changes that occur in circulating immune cells in AD at the single cell level in response to dupilumab treatment. Overall design: After giving informed consent, patients with moderate to severe AD provided whole blood samples prior to starting on therapy with dupilumab. At a subsequent visit (around 4 months after initiating therapy), patients provided a second whole blood sample. PBMCs were isolated from whole blood and the 10X Genomics Chromium platform was used to generate single cell RNA-sequencing libraries, which were sequenced on an Illumina NovaSeq 6000.
