The impact of various fatty acid types on adaptive immunity remains uncertain, and their roles remain unelucidated. In this study, we showed that the enzyme stearoyl-CoA desaturase-1 (Scd-1) and its resulting compound, oleic acid (OA), have a substantial impact on the transformation of CD8+ naïve T cells into effector T cells. Inactivation of Scd-1 triggers the specialization of CD8+ T cells into the Teff subset, enhancing the effector function and mitochondrial metabolism of Teff cells, and OA can partially counteract this. Overall design: To further explore the influence of Scd-1 on the differentiation and functional status of Teffs, we isolated CD8+ T cells from the spleens of both WT and Scd-1-/- mice using MojoSort Mouse CD8 T Cell Isolation Kit (BioLegend), which were then induced to differentiate into Teff cells using IL-2 after three days of stimulation with CD3/28. We then performed gene expression profiling analysis using data obtained from RNA-seq of 2 different cells. Comparative gene expression profiling analysis of RNA-seq data for CD8+ Teff cells from WT and Scd-1-/- mice.
