The strain of Vibrio anguillarum was isolated from the visceral tissue of Takifugu obscurus infected with Vibrio anguillarum. The bacterial species was identified using physiological and biochemical methods and 16s rRNA sequencing. The overnight activated bacterial solution was handed over to Shanghai Shenggong Biotechnology Co., Ltd. for second-generation whole genome sequencing using the HiSeq2500 platform. Use FastQC software to evaluate the quality of the raw data after sequencing, and further filter out low-quality sequences and sequence connectors using Fastp software. Use SPAdes software for genome assembly, remove low-quality contigs (length less than 1000bp or coverage less than 10), and evaluate genome assembly integrity using BUSCO software. Perform gene prediction on the genome using Prokka software, and analyze whole genome features using Rfam, RNAmmer, and tRNAscan SE. Perform functional annotation on the predicted amino acid sequences in the Clusters of Orthologous Groups (COG), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Gene Ontology (GO) databases. Download the whole genome alignment sequence from the NCBI official website, and calculate the average nucleotide identity (ANI) using fastANI software. The reference genome sequence is shown in Appendix 1. Obtain virulence genes through the comparison results of the Virus Factors of Pathogenic Bacteria (VFDB) database. Compare the antibiotic resistance genes carried by bacteria using AMRFinderPlus software and its accompanying drug resistance gene database, Comprehensive Antibiotic Resistance Database (CARD) database.
