We describe a xenograft model for human LCH-like disease in which we transduced human Cord Blood CD34+ HSPCs with a virus coding for BRAF-V600E - the most abundant pathognomonic somatic mutation occuiring in LCH. Cells were subsequently subjected to RNA-sequencing or transplanted into humanized MISTRG or NSG mice. Overall design: CD34+ Cord Blood HSPCs were transduced with a Lentivirus coding for BRAF-V600E and GFP or with a control virus containing a GFP- and a Luciferase contruct instead of BRAF-V600E and were then kept in vitro for 5 days. Samples were then sorted for GFP expression and subjected to bulk RNA sequencing.
