RNAseq was used to identify host and viral transcriptome changes in methionine restricted or SHIN1 treated P3HR-1 cells. Overall design: For methionine restriction, P3HR-1 cells were washed with PBS and then cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.01mM L-methionine for 3 days. Control cells were cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.1mM L-methionine for 3 days. Control or methionine restricted cells were then subjected to RNAseq analysis. For SHIN1 treatment, P3HR-1 cells were treated with 10uM SHIN1 for 3 days. 1mM sodium formate was used to rescue the SHIN1 effects.
